Sera from 77 Japanese patients with chronic hepatitis B virus (HBV) infection were screened for HBV genotypes. They were identified as having genotype B by the single strand conformation polymorphism (SSCP) method, but differed in banding patterns. Genotyping of intragenic sequences by use of hybridization to nested PCR products confirmed that these subjects carried B/d and B/c recombinants. Prion diseases are infectious neurodegenerative diseases that are characterized by the accumulation of an abnormal prion protein, PrPSc, in the central nervous system. The precise mechanism underlying the propagation of PrPSc remains unclear. We studied the mechanism by which PrPSc could spread between neurons in vitro and in vivo. We examined the propagation of PrPSc after the incubation of normal prion protein (PrP(C)) with amyloid-beta (A beta) and its derivatives. PrP(C) was expressed in a neuroblastoma cell line (N2a cells), and the cells were inoculated with PrP(Sc) generated by treating N2a cells with PrP(Sc) isolated from prion-infected mice.
